NEXD: Nanoreactor Bead Extraction of cfDNA for Hypothesis-Free Pathogen Diagnostics

The NEXD project explores new approaches to the molecular diagnosis of infectious diseases that are difficult to detect. At its core is the analysis of cell-free DNA (cfDNA), which enables the identification of pathogens even when classical microbiological methods reach their limits. The project focuses in particular on infections such as endocarditis and invasive pulmonary aspergillosis, where rapid and reliable diagnostics are crucial.
A central aim of NEXD is the development of a novel extraction method based on nanoreactor beads from BLINK AG. These beads enable the efficient enrichment of nucleic acids from large sample volumes. The method is suitable for a wide range of sample types, including plasma, urine, and swabs, and allows for standardized processing even at very low DNA concentrations.
Molecular detection is performed using digital multiplex PCR, which enables sensitive and parallel detection of numerous target sequences. In combination with low-bias sequencing, pathogen profiles can be generated without the need for a predefined diagnostic hypothesis. This enables comprehensive microbiome analysis that includes both known and unexpected pathogens.
In addition, NEXD is developing AI-based software solutions for the evaluation of complex data sets. Automated analysis facilitates interpretation of the results, reduces manual workload, and lowers diagnostic costs. The aim is to establish a method that delivers faster and robust results and can be integrated into routine clinical practice in the long term.
